He-Ne激光照射对淋巴结内巨噬细胞、淋巴细胞影响的实验研究

【摘要】 目的 探讨弱激光对机体免疫刺激作用的机理。 用剂量63.7J/cm2的He-Ne激光照射小鼠神阙穴,照射时间为10天,每天照射5min,(1)采用台盼蓝活体注射显示巨噬细胞吞噬力试验方法;(2)采用吖啶橙(AO)显示核酸的荧光染色法分别观察小鼠肠系膜淋巴结内巨噬细胞的吞噬功能和淋巴细胞内核酸含量的。(3)观察淋巴结组织结构。结果 (1)照射组淋巴结内巨噬细胞吞噬功能较对照组显著增高,差异有非常显著性(P<0.01);(2)照射组淋巴细胞内核酸含量较对照组明显增多。(3)淋巴小结增多增大。结论 适当剂量的He-Ne激光可激活淋巴结免疫功能,提高机体免疫力。 【关键词】 激光;巨噬细胞; 淋巴细胞 An experimental study of He- Ne laser irradiation on the macrophages and lymphocytes in lymph node 【Abstract】 Objective To research the effects of He-Ne laser irradiation on immunity of organism.Methods Shenque acupoints of mice abdomen were irradiated by He-Ne laser at the dosages of 63.7J/cm2 for 10 days,5 minutes each day.n following methods are taken:(1)Observing the phagocytosis of the macrophages in lymph node after trypan blue injecting the live mice.(2)Observing the changes of nucleic acid of lymphocytes with fluorescence microscope.(3) Observing the tissue structure of lymph node by HE stain.Results The four indexes:number of trypan granule,area of granule,ratio of granule area to cell area and granule integral optical density at irradiated group were higher than that of non-irradiated group (P<0.01).The nucleic acid of irradiated groups were higher than non-irradiated group.In addition.the lymphoid nodule in the irradiated groups remarkable changes indicating their activation. Conclusion He-Ne laser at appropriated dosages may activate lymph node and enhance the immunity of organism. 【Key words】 laser;macrophage; lymphocyte 淋巴结是机体的主要周围淋巴器官,位于淋巴循环的通路上,其大小、结构及内含细胞成分的变化与机体的免疫功能状态密切相关。本文采用He-Ne激光照射小鼠神阙穴后,观察激光对肠系膜淋巴结组织结构和其内巨噬细胞吞噬力及淋巴细胞增殖能力的影响。旨在探讨低强度激光对机体免疫刺激作用机制,为临床提供实验依据。 1 材料与方法 1.1 材料 昆明种小白鼠,体重18~22g,雌雄各半,随机分成对照组和照射组,采用上海产1000B(c)He-Ne激光器照射小鼠神阙穴(剪毛),照射功率25mW,照射剂量为63.7J/cm2,光斑直径0.3cm,照射时间为10天,每天照射5min。 1.2 方法 (1)取材前3日,每天往腹腔内注射0.5%的无菌台盼蓝0.3ml,第4日取肠系膜淋巴结。用10%福尔马林固定,常规法制成石蜡切片,用伊红淡然,将切片用HI-CI型医学图像系统于每组600倍光镜下随机选择12屏,统计巨噬细胞吞噬台盼蓝的颗粒数目(GN)、颗粒面积(GA)及其与细胞面积之比(GA/CA)和颗粒积分光密度(IOD) 4项指标,并输入微机作F检验。(2)取肠系膜淋巴结涂片,用Carnoy 氏固定液固定15min,再用0.01% AO缓冲液染色10min,置OLYMPUS BHF-型荧光显微镜的高倍镜下观察,DNA呈黄色荧光,RNA呈红色荧光,DNA或RNA含量越多,黄色或红色越深越亮。(3)常规HE染色方法观察淋巴结的组织结构。 2 结果 2.1 He-Ne激光对淋巴结巨噬细胞吞噬能力的影响 淋巴结的每组切片置图像分析系统的光镜下放大600倍,随机选择12屏观察淋巴结髓质区,巨噬细胞胞质内出现蓝色颗粒为阳性,台盼蓝颗粒呈大小、粗细不等,分布不均,颜色深浅各异。图像分析结果用统计软件作F检验,见表1。
